The in vitro assembly of tubulins from sea-urchin eggs and rat brain: use of heterologous seeds.
نویسندگان
چکیده
Sea-urchin tubulins can be assembled in vitro to form microtubules provided that a suitable seed is present. Although fragments of sea-urchin microtubules fail to act as seeds, since they apparently dissociate, short pieces of rat brain microtubule will act as nucleation sites for the assembly of sea-urchin tubulins. In addition, rat brain tubulins appear to undergo a certain amount of in vitro assembly when seeded with fragments of microtubules derived from seaurchin sperm tails. Assembly on a heterologous seed indicates that the current in vitro assembly conditions may be insufficiently stringent.

Assembly of unfertilized sea urchin egg tubulin at physiological temperatures.
Tubulin was purified from unfertilized eggs of the sea urchin Strongylocentrotus purpuratus by DEAE-column chromatography and cycles of temperature-dependent assembly and disassembly. Tubulin-containing column fractions self-assemble into intact microtubules in the absence of high molecular weight microtubule-associated proteins. Egg microtubules assembled during the third cycle of assembly fol...
متن کاملColchicine-binding activity distinguishes sea urchin egg and outer doublet tubulins
The colchicine-binding activity of tubulin has been utilized to distinguish the tubulins from two distinct microtubule systems of the same species, the sea urchin Strongylocentrotus purpuratus. We have analyzed the colchicine-binding affinities of highly purified tubulins from the unfertilized eggs and from the flagellar outer doublet microtubules by van't Hoff analysis, and have found signific...
متن کاملMultifunctional Ca2+/calmodulin-dependent protein kinase is necessary for nuclear envelope breakdown
The role of multifunctional Ca2+/calmodulin-dependent protein kinase (CaM kinase) in nuclear envelope breakdown (NEB) was investigated in sea urchin eggs. The eggs contain a 56-kD polypeptide which appears to be a homologue of neuronal CaM kinase. For example, it undergoes Ca2+/calmodulin-dependent autophosphorylation that converts it to a Ca2(+)-independent species, a hallmark of multifunction...
متن کاملEmbryonic, larval and juvenile development of tropical sea urchin, Diadema setosum
Diadema setosum (Leske, 1778), is one of the common echinoids widely distributed in the Indo-West Pacific Ocean, where it occurs from the Red Sea, Persian Gulf and the east coast of Africa to Japan, Australia and Malaysia. To investigate the developmental basis of morphological changes in embryos and larvae, we documented the ontogeny of D. setosum in a controlled laboratory condition at the In...
متن کاملSerological Similarity of Flagellar and Mitotic Microtubules
An antiserum to flagellar axonemes from sperm of Arbacia punctulata contains antibodies which react both with intact flagellar outer fibers and with purified tubulin from the outer fibers. Immunodiffusion tests indicate the presence of similar antigenic determinants on outer-fiber tubulins from sperm flagella of five species of sea urchins and a sand dollar, but not a starfish. The antibodies a...
